Brief description of the project AP09259567 "Multi-botanical Oral Supplement to Suppress the Spreading of SARS in Humans"
The strategic policy of the Republic of Kazakhstan is aimed at creating high-tech technologies based on pharmaceutical production by reducing planned dependence on imported medicines, i.e. domestic production forces, raw materials, the scientific and technical potential of the country and high-tech technologies. Today, one of the main directions of development of the Republic of Kazakhstan is the development of the pharmaceutical industry at the expense of domestic raw materials. More than 7,000 medicines have been registered in the State register of the Republic of Kazakhstan. The share of domestic drugs is 30 %, in value-10 %, respectively, 90 % of pharmaceutical products are met by the demand for imported drugs. Providing the pharmaceutical market with new medicines of domestic production in the conditions of import dependence in the country is an important stage in the process of formation of the pharmaceutical industry in Kazakhstan. In this case, it is important to study the development potential of domestic pharmaceutical products by studying the production of a new lead natural product using a wide range of medicinal plants in Kazakhstan.
A number of coronavirus outbreaks have struck the world in recent times (MERS-CoV, SARA-CoV, and SARS-CoV-2) and that it would be reasonable to predict that future outbreaks will occur. Medicinal plants have been the source of various medical product leads in drug discovery. It is not a surprise that plant products and their analogues have could be employed as an early line of defence against SARS. It would be valuable to have botanical preparations that could help prevent/build resilience to future respiratory viral infection outbreaks. The main role of these preparations is to suppress SARS-CoV-2 cell entry through targeting transmembrane protease serine 2 (TMPRSS2) and block coronavirus replication via targeting the viral 3-chymotrypsin-like cysteine protease (3CLpro). Hoffmann et al., (Cell 181 2020) has shown that SARS-CoV-2 cell entry depends on angiotensin-converting enzyme 2 (ACE2) and transmembrane protease serine 2 (TMPRSS2). Targeting ACE2 will present some challenge as it has a beneficial role in many diseases such as hypertension, diabetes, and cardiovascular disease, so targeting TMPRSS2 will become more visible. Medicinal plants have been the source of various medical product leads in drug discovery. It is not a surprise that plant products and their analogues have could be employed as an early line of defence against influenza virus. Several plants are known to possess potential potency towards TMPRSS2 such as; Scutellaria baicalensis, Aloe vera, and Hydrocotyle leucocephala. The edible Hydrocotyle leucocephala that contains hinokinin which has been shown to potentially inhibit TMPRSS2 (RSC Advances 10 (2020).
The use of nutraceutical formula with potent established activity against such types of viruses in a combination of immune-boosting botanical seems to be the quickest approach making benefit of the wealth of information that is already known. The present project for natural drug research from medicinal and edible plant resources will be engaged in cutting edge fundamental and translational research on phytochemical compositions to develop safe and effective phytomedicines of defence against viral influenza, SARS and future human health threatened pandemics. Similar to TMPRSS2 approach, we will confirm the activity of several 3CLpro known active plants in our research center to select the most suitable candidate. Our attempt is to establish the potent and safe botanical formula of fifteen different medicinal and edible plants (1-15). And this recipe has long been used as a Traditional Kazakh Medicinal (TKM) remedy against influenza, inflammation and various viral infections. We have expanded and improved the recipe based on the latest research from other plants. Thus, we present this recipe as a meaningful and advanced effective remedy. Moreover, the multi-botanical formula is being registered at «The Committee for Sanitary and Epidemiological Control of the Ministry of Health of the Republic of Kazakhstan» as an oral supplement in granule form.
The goal research project mainly focused on using sevral selected bioactive medicinal and edible plants, to generated new antiviral botanical oral supplement, determinate its chemical constituents, isolation of the main natural product leads, study their structure and pharmacological activities, run in vitro screening towards TMPRSS2 and 3CLpro to develop new methods and techniques to create natural, safe multi-botanical preparation to suppress the spreading of SARS in humans.
The developed product. Within the framework of the project, a botanical formula was developed, consisting of fifteen types of medicinal and nutritious herbal supplements that have a powerful effect against coronavirus. The main role of these drugs is to suppress the penetration of SARS-CoV-2 cells through targeting transmembrane serine protease 2 (TMPRSS2) and blocking the replication of coronavirus through targeting viral 3-chymotrypsin-like cysteine protease (3CLpro). For the first time, a multi-botanical oral supplement was created that has preventive and curative properties with a main focus on the influenza virus, SARS in Kazakhstan. A safe product from medicinal and edible plants has been created that provides short-term access to the influenza virus and SARS to patients. This formula guarantees its function through two main dual mechanisms, including the inhibition of coronaviruses and the enhancement of the immune system (Коронавирусқа қарсы тұнба – көп аурудың емі // Egemen Qazaqstan, 06.03.2023, https://egemen.kz/article/337077-koronavirusqa-qarsy-tunba-%E2%80%93-kop-aurudynh-emi).
The economic point realization of the project will highlight the new biologically active natural leads needed to obtain drugs with a further introduction to medicine and will contribute to accelerated innovation development of the economy of the Republic of Kazakhstan.
Social effect. The developed technology will open new businesses and jobs, improve profitability and the economic effect of existing enterprises. Young Scientists participation in the project will allow improving the quality of education and training of highly qualified specialists that in the future will increase their ability for a better career development.
The research is carried out at the Research Center for Medicinal Plants (https://farabi.university/science/nii/51) On the basis of the Research Center for Medicinal Plants of the Al-Farabi Kazakh National University, a low-tonnage production of innovative products based on plant raw materials was opened (ҚазҰУ-да жаңа өндіріс іске қосылды // Farabi Media – KazNU, 12.09.2022, https://youtu.be/7Ny1OnRCqrE). Experimental batches of biologically active supplements have been released: antiviral, immunomodulatory, for women's health, and for liver protection. Herbal teas are produced in order to regulate blood pressure; reduce blood sugar levels in diabetes; maintain immunity; maintain the full functioning of the stomach. Work is underway on state registration and declaration of conformity of products.
Collaborations under the frame of the projects: Nazarbayev University, Astana, Kazakhstan; National Center for Natural Products Research, School of Pharmacy, University of Mississippi, Oxford, MS 38677, USA; Division of Applied Life Science (BK21 four), IALS, Gyeongsang National University, Jinju 52828, Republic of Korea, Tianjin Institute of Industrial Biotechnology (TIB), Chinese Academy of Sciences (CAS), and Xinjiang Technical Institute of Physics and Chemistry , CAS.
Based on the project, the Plant expeditions (2021-2023) were carried out with all members of the research team (North, South, East , and Almaty region of Kazakhstan). Based on the project trained young scientists: High school students, Bachelor, Master and PhD students.
Based to the obtained results from present project, our team has been submitted 2 articles in journals which recommended by CQA (Committee for Quality Assurance, Ministry of Education and Science of Republic Kazakhstan) of which, 2 articles in a peer-reviewed scientific journals, meanwhile presented 13 paper presentations at national and international conferences and submitted one patent applications in the Republic Kazakhstan (RK). Moreover, the multi-botanical formula has been tested by accredited laboratory of "NUTRITEST" LLP (No. 23P dated 02/23/2022 for Biologically active additive "TKM-Immuno"), and passed through the expert evaluation of materials for Biologically active additive "TKM-Immuno", LLP "OO Kazakh Academy of Nutrition" (No. 19/EC-105-22 dated 30.03.2022). Successfully the oral supplement has been registered at «The Committee for Sanitary and Epidemiological Control of the Ministry of Health of the Republic of Kazakhstan» as an oral supplement in granule form («TKM-Immuno» KZ.16.01.98.003.R.000318.06.22, 17.06.2022).
